• dATP + dGTP + dCTP + dTTP + ATP + Mg++ + template DNA = NEW DNA!!!

• Nucleotides added to 3’ end at a rate of 650 nucleotides/min. at 37oC

• Problems:

  • In vivo replication rate is 100X faster
  • Incapable of replicating intact DNA
    • (must be nicked)
  • Synthesis only 3’- 5’
  • Exonuclease activity
